HPLC 99JAN 01-08-99 through 02-07-99 Samples taken from rosette Go-Flo bottles and analyzed with HPLC for pigments. year, decimal day, date, latitude (decimal degrees), longitude (decimal degrees), LTER grid line, LTER grid station, irradiance (%), LTER event #, depth (m), chlorophyll c2 (ug/l), peridinin (ug/l), 19'-butanoyloxyfucoxanthin (ug/l), fucoxanthin (ug/l), 19'-hexanoyloxyfucoxanthin (ug/l), diadinoxanthin (ug/l), alloxanthin (ug/l), diatoxanthin (ug/l), lutein (ug/l), zeaxanthin (ug/l), chlorophyll b (ug/l), chlorophyll a (ug/l), alpha carotene (ug/l), beta carotene (ug/l) Water was sampled at all stations with the exception of the Palmer inshore stations F -J, from Go-Flo bottles tripped at the surface, and bottles tripped at depths corresponding to 50, 25, 10, 5, and 1 percent of measured surface PAR. Samples were filtered onto GF/F filters and frozen for a minimum of 24 hours. Pigments were then extracted in 90% acetone for 24 hours at -60 degrees Celsius. Samples were then injected onto a Waters C18 300mm column and separated by HPLC using a method adapted from Wright et al, 1991. Peaks were identified by diode array spectrum analysis and integrated using Hitachi's D-7000 system; concentrations were calculated based on external standard curves in Microsoft Excel. Water sampled from Go-Flo bottles, which corresponded to light levels used in primary productivity simulated in situ experiments (see Sampling Frequency, above). Concentrations based on calibrations done in the 1998-1999 field season, with the exception of lutein, zeaxanthin and beta carotene, which are based on 1997 calibrations. Please note that this data set may be updated as adjustments are made to instrument calibration.
