Samples taken from Go-Flo bottles and analyzed with HPLC for photosynthetic pigments. Samples were filtered onto GF/F filters and frozen at -60 degrees Celcius for a minimum of 24 hours. Pigments were then extracted in 90% acetone for 24 hours at -60 degrees Celsius. Samples were then injected onto a Waters C18 300mm column and separated by HPLC using a method adapted from Wright et al, 1991. Peaks were identified by diode array spectrum analysis and integrated using Hitachi's D-6500 system; concentrations were calculated based on external standard curves in Microsoft Excel. Water sampled from Go-FloS bottles which corresponded to light levels used in primary productivity simulated in situ experiments (see Sampling Frequency, above). lter, antarctic, HPLC, pigments ASCII, comma separated 95crpigs Maria Vernet Wendy Kozlowski Maria Vernet Wendy Kozlowski, Ted Ruel, Stewert Lamerdin, John Harrison, Karen Patterson, Christy Campbell Wendy Kozlowski, Stewert Lamerdin Wendy Kozlowski Wendy Kozlowski Wendy Kozlowski 12-31-96 09-25-97 Wright, S.W., S.W.
