To determine photosynthesis-irradiance (P-I) relationships, twenty- five 1 ml whole water samples for each discrete field sample were inoculated with H14CO3 (final concentration 3-6 mCi/ml) and placed in newly designed photosynthetrons. The photosynthetrons consisted of a milled aluminum block holding fifty 7 ml scintillation vials. Each block had hollowed out channels which circulate an ethylene glycol:water (1:1) solution for temperature control (+/- 0.05oC). The light source was two 75 watt FOS-8 Cool-Lux Inc. lamps (one lamp for each set of 25 samples). In order to eliminate the spectral bias of carbon uptake in white light, light was passed through a CuSo4 filter to eliminate far-red wavelengths of light. Neutral density Plexiglas filters were used to produce a variety of light intensities ranging from 0 to 1300 uEm-2s-1. After a 1.5 hour incubation at in situ temperatures , samples were fixed with 20 uL formalin solution, acidified with glacial acetic acid:methanol (1:30), and heat dried. National Diagnostics Liquiscint scintillation fluor was added and samples were counted on a LKB 1217 scintillation counter.
